BINNION, PETER F., JAMES 0. DAVIS, TORREY C. BROWN, AND MICHAEL J. OLICHNEY. Mechanisms regulating aldosterone secretion during sodium depletion. Am. J. Physiol. 208(4) : 655-661. rg65.LSteroid secretion was studied in conscious dogs before and during 4 days of Na depletion. A threefold increase in aldosterone secretion occurred but corticosterone output was unchanged. Plasma renin was markedly elevated. Plasma Na and K concentrations were unchanged until the 4th day of Na depletion at which time plasma Na was decreased and plasma K was elevated. The Na content of the adrenal cortex expressed as fat-free tissue solids was decreased. It seems likely that the adrenocortical Na loss was extracellular; adrenocortical K content was unaltered. It is suggested that the normal rate of corticosterone secretion during Na depletion is maintained by a negative corticosteroid feedback mechanism, since corticosterone output was consistently increased in Na-depleted hypophysectomized dogs. Aldosterone secretion was three times as great in the presence as in the absence of the anterior pituitary during Na depletion. It is concluded that increased activity of the renin-angiotensin system is the primary mechanism leading to hyperaldosteronism during Na depletion and that the adenohypophysis plays an important supportive role.

Renin and Aldosterone Secretion in Man
In six normal volunteers maintained on a constant dietary intake, serial studies were made of plasma renin activity, blood volume, aldosterone secretion and sodium balance, first following dietary and diuretic-induced depletion of sodium and blood volume, then following a period of sodium repletion during a maintained reduction in blood volume, and finally after blood volume repletion. In the f...
متن کاملRelative influence of acute sodium and volume depletion on aldosterone secretion in nephrectomized man.
Plasma aldosterone concentration can increase in anephric man during hemodialysis despite the lack of kidneys to produce renin. Because the metabolic clearance rate of aldosterone does not change during hemodialysis, aldosterone secretion must increase in response to sodium depletion, volume depletion, or both. This study was designed to determine the relative influence of sodium and volume dep...
متن کاملRenal origin of an aldosterone-stimulating hormone in dogs with thoracic caval constriction and in sodium-depleted dogs.
Recent studies (1-5) indicate that the immediate stimulus to aldosterone production is humoral. Evidence for secretion of an aldosterone-stimulating hormone (ASH) by the kidney in response to acute blood loss was presented at the Laurentian Hormone Conference in 1960 (4). The present study is concerned with the locus of secretion of an ASH in two other experimental situations: 1) hyperaldostero...
متن کاملSites of action of sodium depletion on aldosterone biosynthesis in the dog.
Secretion of cortisol, corticosterone, and aldosterone was measured in vivo in normal and sodium-depleted hypophysectomized dogs. Biogenesis of steroids was then measured in vitro with outer slices of the adrenals of the same dogs. In some studies, metyrapone or puromycin was added. In vivo, sodium depletion stimulated the production of cortisol, corticosterone, and aldosterone. In vitro, tissu...
متن کاملSensitization of the Adrenal Cortex to Angiotensin II
The effect of sodium depletion on the dose-response relationships of angiotensin II to aldosterone and blood pressure was studied. Arterial plasma angiotensin II and aldosterone and arterial blood pressure were measured before and during the incremental infusion of angiotensin II into sodium-replete and sodium-deplete subjects. Sodium depletion caused a distinct steepening of the angiotensin H-...
